Our TMC Research 486DX PC recently started having a keyboard problem at bootup. It gives 2 beeps (AMIBIOS POST error) and "Keyboard error", or sometimes "KB/Interface error" on the monitor. Sometimes Windows will actually load, but without keyboard input. The serial port mouse began acting wierd i.e., lack of control, around the same time.

I already tried two spare keyboards with no effect. Should I suspect a motherboard or keyboard controller failure, maybe? Is there a way to repair this without expensive diagnostic equipment?

Possibly it is the keyboard controller and on a 486 mobo it is problably more expensive to get the controller replaced than to get a new mobo. If you are set on keeping the 486 and not upgrading to a newer machine then you can try here for a replacement:CSO (http://www.computersurplusoutlet.com/), a 486DX-66 for about $20 US. To get someone to replace the keyboard controller would be a minimum of $35 (about the average for 1 hour in the shop) plus the price of the replacement part.
