Not having any definition, but beeing in the posting mood, here comes my educated estimate:
Tweaking is setting up your machine in such a way that you take maximum advantage of the computational power at your discretion. This may be things like not running any unnessecairy programs in the background, but also hardwarestuff like setting the jumpers on your mobo of vidcard so and so to gain maximum advantage of each components caracteristics.
At a certain point the tweaking stops and the overclocking begins. This is when you start getting ore Mhz out of a component than is labeled on the box.
But remember: this is my educated guess at it, I might be wrong have it all wrong.

I think it can be compared to fine tuning a car--make a little adjustment here--a little adjustment there.

I run WinME on my main computer. As soon as I do a fresh install, I delete the folder of Internet providers that M$ always insists on istalling. I then go into msconfig and turn off Task Scheduler and a number of other programs I don't use. I go into WinME settings for Win Explorer and set it to show all files and extensions. I put the view back to the old version rather than the graphics heavy web version. I adjust the Restore cache size to reduce the space it occupies. And so on and so on and so on.
There are utilities I might install to do certain things efficiently and others I disable to keep them from slowing things down. Many programs that I install involve similar tweaking, like browsers, antivirus, firewall and others. If you let most programs do whatever they want pretty soon you can't find your desktop for all the clutter and your RAM will be used by all sorts of background programs that you don't need 99% of the time. Your system will crash more often and run slower.

Verrrry carefully. Here's a place you can go http://www.dougknox.com/index.html to get some tweaks. Be very careful though. A lot of these tweaks has to do with making changes to the Registry. I would not advise that unless you are familiar working with the Registry. If you are, then you would know that the Registry should be backed up before messing with it.

Not to worry:

You have also stated that you are not comfortable making manual
changes to your registry at this time. Try this site to download
a trial version of winguides tweak manager. Try it, see if you
like it. It's works with all versions of Windows:
